BOSTON, May 14: Richard Slayman, the first patient to receive a genetically
modified pig kidney, has died two months after the xenotransplantation at Massachusetts
General Hospital in Boston, it was announced last Saturday.
The four-hour surgery performed on March 16 marked a major milestone in the
quest to provide more readily available organs to patients, the hospital had
said.
As animal-to-human transplants are crucial to ease the huge organ shortage,
Slayman's case has been hailed as a historic milestone. As human immune system
destroys foreign animal tissue, a genetically modified pig kidney was transplanted
on Slayman.
The hospital said there is “no indication” that Slayman’s death “was the result
of his recent transplant.”
There have also been transplants of other pig organs. Maryland surgeons transplanted
a pig heart on a dying patient but he survived only two months.
Much before Slayman's case, a genetically modified pig kidney was transplanted
on one Maurice Miller, 57, who had cancer and was brain dead, at NYU Langone
in New York City. He died on September 12, 2023, after 61 days. The pig kidney
worked normally inside his brain-dead body. Alongwith the modified pig kidney,
the thymus was also transplanted, . beneath the kidney. That was to prevent
the transplanted kidney from being attacked by the human immune system by learning
to recognize the pig antigens as its own.
In another case, the University of Alabama at Birmingham found two gene-edited
pig kidneys functioned for seven days in a brain-dead recipient. It also used
"immunosuppression drugs."
Slayman, 62, a resident of Weymouth, near Boston. had been living with Type
2 diabetes and hypertension for many years. He was on dialysis for many years
before receiving a kidney transplant from a human deceased donor in December
2018 at Massachusetts General Hospital. It worked for five years and he was
back to dialysis in May 2023.
He underwent the pig kidney transplantation surgery on March 16. It took six
hours. The pig's kidney had received 69 edits to prevent rejection by the patient's
immune system. He was quite happy when he left the hospital on April 2 that
he was no more required to undergo dialysis - “free from the burden of dialysis
that has affected my quality of life for many years.”
The pig kidney was provided by eGenesis, a pharmaceutical company, from a pig
donor genetically-edited using CRISPR-Cas9 technology. Certain pig genes were
removed and some human genes were added.
The hospital said that Slayman “will forever be seen as a beacon of hope to
countless transplant patients worldwide.”
There are over a lakh patients waiting for kidney transplant in the US.
